Mechanism of action

GANATON stimulates GI motility. Itopride hydrochloride enhances propulsive motility of the gastrointestinal tract due to antagonism with dopamine D2-receptors and dose-dependent inhibition of acetylcholinesterase activity. Itopide activates the release of acetylcholine and inhibits its destruction.

Itopride hydrochloride has a specific effect on the upper GI tract, accelerates the transit through the stomach and improves its emptying.

The drug also has an antiemetic effect due to interaction with D2-receptors located in the trigger zone. Itopride caused dose-dependent suppression of vomiting caused by apomorphine.

Itopride hydrochloride does not affect serum levels of gastrin.

Indications and usage

Relief of functional non-ulcer dyspepsia (chronic gastritis), including:

- abdominal distention;

- feelings of rapid satiety;

- pain or discomfort in the upper abdomen;

- anorexia;

- heartburn;

- nausea;

- vomiting.

Contraindications

- Gastrointestinal bleeding, mechanical obstruction or gastrointestinal perforation;

- pregnancy;

- lactation period (breastfeeding);

- children's age up to 16 years;

- Hypersensitivity to itoprid or any auxiliary component of the drug.

Dosage and administration

appoint inside 50 mg (1 tablet) 3 times / day. before meals. The recommended daily intake is 150 mg. This dose may be reduced, taking into account the age of the patient.

Adverse reactions

From the hemopoietic system: leukopenia, thrombocytopenia.

On the part of the endocrine system: increased levels of prolactin, gynecomastia.

From the side of the central nervous system: dizziness, headache, tremor.

From the digestive system: diarrhea, constipation, abdominal pain, increased salivation, nausea, jaundice.

Allergic reactions: skin flushing, itching, rash, anaphylaxis.

Changes in laboratory parameters: increased activity of AST and ALT, GGT, ALP and bilirubin level.

Metabolic interaction is hardly possible, because Itoprid is metabolized by the action of flavin-dependent monooxygenase, and not the isoenzymes of the cytochrome P450 system.

With simultaneous use Ganatona with Warfarin, diazepam, Diclofenac sodium, ticlopidine hydrochloride, Nifedipine and nicardipine hydrochloride there were no changes in itopride binding to proteins.

Itoprid increases the motility of the stomach, so it can affect the absorption of other simultaneously used inside drugs. Particular care should be exercised when using drugs with a low therapeutic index, as well as forms with a slow release of the active substance or drugs with an enteric coating.

Anti-ulcer agents such as cimetidine, Ranitidine, teprenone and cetraxate do not affect the prokinetic effect of itoprid.
