Mechanism of action

Delagil - antiprotozoal agent, also has an immunosuppressive and anti-inflammatory effect. Causes the death of asexual erythrocyte forms of all types of plasmodia. It has a gametocidal effect, with the exception of Plasmodium falciparum (exhibits anti-gametocidal action). Due to inhibition of the synthesis of nucleic acids, it has a moderately pronounced immunosuppressive and nonspecific anti-inflammatory effect.

Indications and usage

Malaria (prevention and treatment of all types), extraintestinal amebiasis, amebic liver abscess, SLE (chronic and subacute forms), rheumatoid arthritis, scleroderma, photodermatosis, late skin porphyria.

Contraindications

Hypersensitivity, hepatic and / or renal failure, inhibition of bone marrow hematopoiesis, severe rhythm disturbances, psoriatic arthritis, neutropenia, porphyrinuria, pregnancy. With caution. Deficiency of glucose-6-phosphate dehydrogenase, retinopathy, epilepsy.

Dosage and administration

Delagil take inside. For the prevention of malaria: 500 mg in the first week, 2 times, then 1 time on the same day of the week.Malaria treatment is carried out according to the following scheme: 1st day - once 1 g, after 6–8 h - 500 mg; on the 2nd and 3rd day of treatment - 750 mg daily in one session. In amebiasis: 500 mg 3 times a day for 7 days, then 250 mg 3 times a day for another 7 days, then 750 mg 2 times a week for 2-6 months. In rheumatoid arthritis: 250 mg 2 times a day for 7 days, then 250 mg daily for 12 months. With systemic lupus erythematosus: 250–500 mg daily. With photodermatosis: 250 mg daily for 1 week, then 500-750 mg weekly.

Adverse Effects

On the part of the digestive system: nausea, vomiting, gastralgia. Nervous system disorders: dizziness, headache, sleep disturbances, psychosis, seizures. On the part of the cardiovascular system: myocardial damage with changes on the ECG, lowering blood pressure. On the part of the senses: with prolonged use - corneal clouding, retinal damage, blurred vision, tinnitus. Allergic reactions: dermatitis, photosensitivity. Other: myalgia, leukopenia, discoloration of the skin and hair.

Special notes

During the period of treatment, it is necessary to conduct systematic examinations of an oculist and general blood tests.

Overdosage

Symptoms: vomiting, impaired consciousness, visual disturbance, convulsions, collapse, depression of the respiratory center, even death (within 2 hours from the moment of respiratory depression).

Treatment: gastric lavage, ingestion of Activated carbon at a dose of 5 times the estimated dose of chloroquine; challenge vomiting, peritoneal dialysis, plasmapheresis.
