Mechanism of action
GENELE is a synthetic steroid antiprogestogen drug. It blocks the action of Progesterone at the receptor level, does not possess gestagen activity. Antagonism with GCS due to competition at the level of communication with receptors was noted.
Depending on the phase of the menstrual cycle, it causes an increase in the contractility of the myometrium, stimulating the release of interleukin-8 in choriodecidual cells, increasing the sensitivity of the myometrium to prostaglandins. As a result of the action of the drug, desquamation of the decidual membrane and removal of the ovum occurs. It causes inhibition of ovulation, changes in the endometrium and prevents the implantation of a fertilized egg.
Emergency (post-coital) contraception within 72 hours after intercourse without prior use of contraceptives or methods, as well as their unsuccessful use (including an error in using the calendar method, unsuccessful interruption of sexual intercourse, breaking or slipping of the condom).
Contraindications
- acute or chronic renal and / or liver failure;
- long-term therapy with glucocorticosteroid drugs;
- porphyria;
- anemia;
- violations of hemostasis (including previous treatment with anticoagulants);
- The presence of severe extragenital pathology;
- adrenal insufficiency;
- pregnancy;
- lactation period;
- a history of hypersensitivity to mifepristone and / or auxiliary components.
Dosage and administration
Take 1 pill inside 2 hours before or 2 hours after a meal (within the next 72 hours after unprotected intercourse), regardless of the phase of the menstrual cycle.
Adverse reactions
From the reproductive system: bleeding from the genital tract, discomfort and pain in the lower abdomen, exacerbation of inflammatory diseases of the uterus and appendages.
Gastrointestinal: nausea, vomiting, diarrhea.
From the side of the central nervous system: headache, dizziness.
Other: weakness, hyperthermia, urticaria.
