pharmachologic effect

Solian has a neuroleptic, antipsychotic effect.

Indications

Acute and chronic schizophrenia, accompanied by pronounced productive (delusions, hallucinations, mental disorders) and / or negative (affective flattening, lack of emotionality and withdrawal from communication) disorders, including in patients with a predominance of negative symptoms.

Contraindications

Hypersensitivity to the active substance or to other components of Solian; concomitant prolactin-dependent tumors, for example, pituitary prolactinoma and breast cancer; pheochromocytoma; children's age up to 14 years; breast-feeding; combination with drugs that may contribute to the development of atrial fibrillation (quinidine, disopyramide, Amiodarone, sotalol), as well as with bepridil, cisapride, sultopridom, thioridazine, Erythromycin, vincamine, halofantrin, pentamidine, sparfloxacin; combination with levodopa.

With care - pregnancy, epilepsy, parkinsonism, old age, renal failure.

Side effects

Frequent (5-10%): insomnia, anxiety, agitation.

Less frequent (0.1-5%): drowsiness, gastrointestinal disorders such as constipation, nausea, vomiting, dry mouth.

Like other neuroleptics, amisulpride causes an increase in the level of prolactin in the blood plasma, which returns to its previous value after the withdrawal of Solian. It can cause galactorrhea, amenorrhea, gynecomastia, chest pain, impotence, as well as weight gain.

Acute dystonia (spastic torticollis, ocular crises, trismism) may develop, which is reversible and corrected with anti-Parkinsonian drugs. The frequency of extrapyramidal symptoms, which depend on the dose, remains very low in the treatment of patients with a predominance of negative symptoms in doses of 50-300 mg / day.

Reports of tardive dyskinesia, characterized by rhythmic, involuntary movements mainly of the tongue and / or face, usually refer to cases of prolonged use of the drug. Parkinsonian drugs are not effective, they can cause worsening of symptoms.

In rare cases, arterial hypotension and bradycardia may develop, as well as prolongation of the QT interval, and very rarely atrial fibrillation.

Allergic reactions, elevated levels of liver enzymes, mainly transaminases, and cases of convulsive seizures are rarely reported.

Dosage and administration

Solian is taken orally. Recommended use at a dose of 400 to 800 mg / day. In some cases, the daily dose may be increased to 1200 mg. Doses should be increased taking into account individual tolerance. The safety of doses above 1200 mg / day has not been adequately investigated, so they should not be used.
